During the wave you should Tower boost stack, HB Monk auto shooting at west crystal behind the physical beam, sit holo barb by spiderqueen spawn to generate clones, help DPS on 4th char to speed up wave.
Spiderqueen boss fight HB Monk from the platform (above forge) auto shooting at spider queen spawn, 2nd DPS char auto shooting at spider queen spawn, Tower boost Stack, and shred the Spiderqueen boss with holo clones. The boss should die after 2 hawk stances.
